Samuel Cahoon was the eldest child born to William Colquhoun and Deliverance Peck. Samuel's father was killed by Indians when he was only 12.

Samuel married Mary Hunter, a girl 16 years his junior, in 1698. They left New England for Virginia shortly thereafter and settled in a place that would later bear the family name, Cahoons Creek. Samuel and Mary had six children, the last was born several months after Samuel's death in 1704 at the age of 41. Mary outlived her husband by only 7 years.

Some sources suggest that Samuel was either a shoe maker or shoe salesman. Another source says he had a court fight with a Richard Wildinson.

Cahoons Creek today lies within the city limits of Suffolk, Virginia, about 5 miles west of downtown.
